This is a full-time fixed term 3 year position on Crick Terms and Conditions of Employment.

OVERVIEW

We are looking for a highly motivated Senior Lab Research Scientist to join the laboratory of Kathy Niakan, to understand mechanisms of lineage specification in human embryos and stem cells. A successful candidate will support a team of 4 PhD students and postdocs. The majority of their time will be spend in the human embryo and stem cell facility where they will support multiple tasks including the maintenance of lab equipment, the organisation and culture of embryos, genome editing, advanced stem cell derivation and culture. In addition, the post holder will assist with the coordination of research licences, lab reagents and the lab budget.

 

The successful candidate is likely to be an energetic, focused, and productive individual with a desire to work in a congenial, dynamic, and collaborative research environment. Good organizational, analytical, and communication skills are essential.

 

The Laboratory Research Scientist will be a member of the Niakan lab at the Francis Crick institute at the Mill Hill Laboratory. The lab will move to the Francis Crick institute central London site around September 2016. Working alongside other laboratory members, the post holder will support multiple research projects, enabling the accurate and full interpretation of data generated both externally and within the lab. The post holder will report directly to the Group Leader, Kathy Niakan.

 

PROJECT SUMMARY

The focus of our lab is to understand the influence of signalling and transcription factors on differentiation during early human development. During preimplantation development totipotent human zygotes differentiate into pluripotent embryonic cells, which form the foetus, and extra-embryonic cells, which form the placenta and yolk sac. The central question we are addressing is what are the molecular mechanisms that regulate embryonic stem cell pluripotency and how is it disengaged during cellular differentiation? We seek to define the genetic hierarchy acting during differentiation, the influence of extracellular signalling and the extent to which these mechanisms are conserved between humans and mice. The molecular basis of these early cell lineage decisions are of fundamental biological importance and have significant clinical implications for infertility, miscarriages, developmental disorders and therapeutic application of stem cells.

 

Our lab uses approaches of retroviral transduction to derive human induced pluripotent stem cells; mouse embryonic and extraembryonic stem cell derivation and genome editing    to generate genetically modified embryos and stem cells; microdissection/micromanipulation and computational biology to profile human preimplantation embryos and novel culture conditions to derive alternative human stem cell lines. Our lab has a Human Fertilisation and Embryology Authority (HFEA) research licence and we collaborate closely with in vitro fertilisation clinics.

KEY RESPONSIBILITIES

These include but not limited to;

  • To maintain lab and cell culture equipment and ensure that the lab and cell culture activities run as efficiently as possible
  • To help manage the lab consumables budget and order as appropriate
  • Liaise with other support staff as appropriate
  • Manage the inventory of embryos, cell lines and animal stocks
  • Assist in the coordination of research licences and lab inspections
  • Conduct experimental work including embryo thaw, genome editing and culture in line with best practice and UK regulation
  • Keep accurate records of all work carried out
  • Problem solve regarding defective equipment
  • Instruct and advise students, post-doctoral fellows, and collaborators on the use of equipment and techniques
  • Coordinate the shipment of reagents to colleagues
  • Assist with and conduct cell culture experiments
  • Attend group meetings and key talks relevant to the lab
  • To communicate the lab results at meetings as appropriate
  • To contribute to the preparation of scientific manuscripts
